New signing Geert de Vlieger - making only his third City appearance - is set to miss the entire season after rupturing his Achilles tendon. Deep into the second half, the Belgian keeper strecthed for a high ball but fell awkwardly and was stretchered off.
David Sommeil will also have a spell on the sidelines after suffering a calf strain, which could keep him out of action for up to five weeks.
Meanwhile, Danny Mills failed a late fitness test and didn't feature but should be fit to fill Sommeil's right-back role at Reading on Monday.
Mikkel Bischoff starred alongside Sommeil in the City defence, replacing Richard Dunne. Of the side that faced Hull, Robbie Fowler, Jon Macken and Claudio Reyna were also left out with Joey Barton, Steve McManaman, and Paulo Wanchope coming into the starting line-up.
And it was Wanchope - partnered by Antoine Sibierski - who had the first City chance, heading over a Shaun Wright-Phillips cross. The England hopeful then struck the post with a dipping volley.
But the hosts took a 14th minute lead when Kenny Miller was allowed enough room to turn a right-wing cross in off de Vlieger's left-hand post.
On 34 minutes City were level though, with Wolves defender Adrian Iencsi chesting Sibierski's cross into his own net as the Wolves keeper was ready to collect behind him.
City were then in front moments before the break when Wright-Phillips hit a tremendous individual goal - his third in three games. He picked up a clearance from substitute Dunne before embarking on a 60-yard run down the right flank, beating one opponent, and firing in right-footed off the bar.
The finish was reminiscent of his derby goal at the City of Manchester Stadium, and was further proof that he is still on top form after a string of impressive performances last term.
McManaman continued to pull the strings in midfield after the break while Bischoff blocked Dean Sturridge when the Wolves striker was almost through.
But Wanchope completed the scoring on 65 minutes after exchanging passes with Sibierski, who brilliantly played the Costa Rican in to score from eight yards.
